摘要

Abstract

Objective To study the effects of DNA methylation inhibitor zebularine on the proliferation,migration and high mobility group box-1 (HMGB1) mRNA expression in ECC-1 cell line(human endometrial carcinoma cell line).Methods MTT method and wound healing assay were used to evaluate the proliferation and migration level of ECC-1.The mRNA expression of HMGB1 was evaluated by Real-time PCR.Results After treatment with zebularine at a final concentration of 200 μmol/L for 24 h,the proliferation and migration of ECC-1 cell lines were significantly inhibited,the absorbance value of MTT method was 0.46±0.04 in theexperiment group,which was significantly lower than that in the control group(0.64± 0.05),(P<0.05);the wound healing rate was 45.23%±3.45% in the experiment group,which was significantly lower than that in the control group(87.45%±6.05%),(P<0.05);the relative mRNA expression of HMGB1 was 1.02±0.10 in control group,and which was significantly higher than that in experiment group(0.25±0.06),(P<0.05).Conclusion DNA methylation inhibitor zebularine could effectively inhibit the proliferation and migration of ECC-1 cell lines,and the effects may re late with the decreasing of HMGB1 mRNA expression.
