Detection of the Expression of Glycan Chain on the Cell Membrane of Gastric Cancer Cells by Lectin Array

Abstract
Objective To detect the distribution pattern of glycan chains on the cell membrane by lectin array and analyze the correlation between differential glycans and the invasion/metastasis of gastric cancer cells (AGS,SGC-7901).Methods Cells were collected with trypsin digestion and labeled with a fluorescent labeling agent.Fluorescence-labeled cells were thencaptured by lectin sites on the chip based on its binding affinity to specific glycan chains.A confocal Confocal laser scanner was used to detect the expression of cell surface glycan chains.Lectin flow cytometry was used for validation.Cell invasion andmetastasis were detected by transwell and wound healing assays.Resuts No significant difference was found in fluorescent signals onmost lectin sites between AGS and SGC-7901 cells.Compared with SGC-7901 cells,LEL and MAL Ⅱ signals were stronger in AGS cells.Mannose,sialic acid,N-acetylglucosamine/glucose,and N-acetylgalactosamine/galactose were expressed in these two kinds of cells.Polylactosamine and α2-3 sialic acid were highly expressed in AGS cells.The ability of invasion and metastasis of AGS cells was stronger than that of SGC-7901 cells.Conclusion Gastric cancer cells with high invasion/metastatic potential express the characteristic polylactosamine and cα2-3 sialic acid.The lectin array-based detection of cell membrane glycan profile will help to screen cancer-related glycan markers thereby assisting the diagnosis and treatment of cancer.关键词
