固定化肾上腺素受体色谱模型的建立及表征
Establishment and Characterization of Immobilized Epinephrine Receptor Chromatographic Model

Abstract
Objective The receptor is an important functional protein in the human body,and acts as the target of many drugs.The receptor is to be constructed by gene recombination and fixed on the carrier to characterize the receptor chromatography,thereby investigating the interaction between receptor and drug and screening the active components.Method The his tag labelled beta2-adrenaline receptor was constructed through gene recombination.The receptor protein was purified by nickel ion chelating column and dialysis.A non covalent method was used to reversibly bond the receptor protein on the macroporous silica gel surface,so as to build receptor chromatography.Results The retention of five types of tool drugs has been determined on the receptor chromatographic column,showing ideal chromatographic peaks with reasonable retention time compared to control.Conclusion The obtained receptor chromatography demonstrated good specificity and stability.It can be used to study the interration between drug and receptor and to screen the active ingredients of target drugs.关键词
