硫代巴比妥酸法测定鱼体肌肉中脂质过氧化物含量的方法优化
Improvement of Thiobarbituric Acid Reaction for the Dertermination of Lipid Peroxidation in the Muscles of Fish
摘要
Abstract
[Objective] To improve the optimum condition for measuring the lipid peroxidation (LPO) in the muscles of fish.[Method] The LPO was determined using spectrophotometer with thiobarbituric acid.The orthogonal design with four factors and three levels was conducted to evaluate the level of LPO in the cyprinoid fish.Four factors were included the gradient of acetic acid,sodium dodecyl sulfate,time and temperature,respectively.[Result] The weight sequence was known as SDS,the gradient of acetic acid,temperature,time by comparing the difference of each factor.The results indicated that the optimum group was A3B1C3D2 by visual analysis and variance analys,namely 20% acetic acid,65 ℃ water bath,60 min.[Conclusion] The study can support the research on the demage of oxidative stress in fish.关键词
