Abstract
Objective To observe the effect of Livin gene in the proliferation and apoptosis in oral squamous cell carcinoma (OSCC) cell line TCA8113.Methods Over expression plasmid pcDNA3.1-Livin and RNAi vector pSuper-shLivin were constructed and transfected into OSCC cell line TCA8113 with lipofectamine 2000.Quantitative PCR or Western blot were used to validate the mRNA or protein expression level.The proliferation rate of TCA8113 cells was detected by MTT assay.The apoptosis was analyzed by Annexin V/PI staining and flow cytometry.Results Livin was highly expressed after pcDNA3.1-Livin transfection.While mRNA level of Livin was silenced after pSuper-shLivin transfection.Over-expression of Livin promoted TCA8113 cells proliferation and inhibited its apoptosis.In contrast,RNA silencing of Livin couldn′t inhibit TCA8113 cells proliferation and promote apoptosis.Conclusion Livin gene can effectively promote the proliferation and inhibit the apoptosis of OSCC cell line TCA8113,and RNA silencing of Livin can effectively enhance the sensitivity of cisplatin-induced apoptosis.关键词
