Abstract

Objective To observe the effects of Nesfatin-1 on the excitability of glucose-sensitive neurons and feeding in the lateral parabrachial nucleus (LPBN),and explore the possible mechanism of Nesfatin-1 on feeding regulation.Methods Wistar rats were randomly divided into Nesfatin-1 group and normal saline (NS) group.Nocturnal food intake was measured by metabolic cage.Applying in vivo electrophysiological technique,the neurons were microinjection with glucose in LPBN to identify if they were glucose-sensitive neurons.Nesfatin-1 and NS were microinjected into glucose-sensitive neurons in LPBN to observe the impact of Nesfatin-1 and NS on neurons in LPBN.Results In Nesfatin-1 group,the six-hour and 12 hour cumulative food intake was lower than that in NS group (t =3.22,3.48;P<0.05),and the difference of that in one-hour and four-hour food intake between the two groups was not significant (P>0.05).There were glucose-sensitive neurons in LPBN,compared with NS group,Nesfatin-1 could significantly excite the glucose-inhibitory neurons in the LPBN (t=-4.99,P<0.01).Conclusion Injection of Nesfatin-1 into lateral parabrachial nucleus can modulate the excitability of glucose-sensitive neurons in the nucleus and inhibit the nocturnal feeding in rats.
