早期糖尿病肾病病人RIP140与炎症因子的关系
RELATIONSHIP BETWEEN RIP140 AND INFLAMMATORY FACTORS IN EARLY DIABETIC NEPHROPATHY

Abstract
Objective To explore the relationships between receptor interacting protein 140 (RIP140) and inflammatory factors in peripheral blood mononuclear cells (PBMCs) of patients with early type 2 diabetic nephropathy (DN).Methods This study consisted of 105 patients with type 2 diabetes (T2DM),among them 50 with simple diabetes (DM group) and 55 with early DN (DN group).A normal control group (CN group) consisted of 37 healthy people.Employing enzyme-linked immunosorbent assay (ELISA),levels of serum TNF-α,IL-6 and IL-1β,in each group,were measured,and using RT-PCR method,the expressions of RIP140,TNF-α,IL 6 and IL-1β in peripheral blood mononuclear cells were detected.Results The expression level of RIP140 mRNA in DM and DN groups was higher than that in NC group,and that in DN was higher than that in DM group,the difference was statistically significant (F=13.275,P＜0.05).Compared with NC group,the mRNA levels of RIP140,TNFα,IL-6 and IL-1β in PBMCs and the concentrations of serum TNF-α,IL-6,IL-1β in DN and DM groups elevated.Compared with DM group,the expressions of the above indicators in DN group were increased (F=12.104-20.417,P＜0.05).In the patients with T2DM,the expression level of RIP140 mRNA was positively correlated with the concentration of serum TNF-α,IL-6 and IL-1β,as well as the expressions of TNF-α and IL 6 mRNA (r =0.203-0.414,P ＜ 0.05).Single factor Logistic regression analysis showed that RIP140 mRNA,TNF-α mRNA,IL-1β mRNA,serum TNF-α and IL6 were risk factors for DN.Conclusion In patients with early type 2 diabetic nephropathy,the expression of RIP140-in PBMCs-increases and closely correlated with the expression levels of TNF-α,IL-6,and IL-1β.关键词
