调节小分子GTP结合蛋白Rab27表达对人膀胱癌细胞Cyclin、p-FAK、p-IκB表达的影响
Effect of regulating expression of small molecule GTP binding protein Rab27 on Cyclin, p-FAK and p-IκB expression of human bladder cancer cells

Abstract
Objective To observe the effect of regulating the expression of small molecule GTP binding protein Rab27 on the cell cycle protein (Cyclin),phosphorylated focal adhesion kinase antibody (p-FAK) and nuclear factor κB (pIκB) of human bladder cancer cells.Methods Rab27 plasmid and Rab27 siRNA were transfected into BIU-87 and 5637 cells in logarithmic phase,respectively.The levels of Rab27,Cyclin D1,Cyclin E,p-FAK (Tyr925),p-FAK (Tyr576/ 577) and p-IκB were analyzed by Western blotting in the bladder cancer BIU-87 and 5637 cells before and after transfection.Results Compared with the Rab27 plasmid transfected BIU-87 cells before transfection,the relative expression of Rab27 was increased,and the expression of Cyclin D1,Cyclin E,p-FAK (Tyr925),p-FAK (Tyr576/577) and p-IκB was increased (all P < 0.05).After the Rab27 interference plasmid was transfected into 5637 cells,the relative expression of Rab27 was decreased (all P < 0.05),and Cyclin D1,Cyclin E,p-FAK (Tyr925),p-FAK (Tyr576/577) and p-IκB relative expression was decreased (all P <0.05).Conclusion After the Rab27 plasmid is transfected into BIU-87 cells,the expression of Cyclin D1,Cyclin E,p-FAK (Tyr925),p-FAK (Tyr576/577) and p-IκB is increased.After the Rab27 interference plasmid is transfected into 5637 cells,the expression of Cyclin D1,Cyclin E,p-FAK (Tyr925),p-FAK (Tyr576/577) and p-IκB is decreased.Rab27 may affect the cell cycle of bladder cancer by regulating the expression of NF-κB and FAK signaling pathway.关键词
