Establishment of a mouse colorectal cancer CT26 cell line with stable interference of WTX gene

Abstract
Objective Using a WTX gene recombinant lentivirus interference vector to infect the mouse colorectal cancer CT26 cell hne to establish a mouse colorectal cancer CT26 cell line with stable interference of WTX gene.Methods The recombinant lentiviral was transformed in mouse colorectal cancer CT26 cells,and fluorescence microscopy was used to observe the preliminary infection efficiency.The stably transfected ceils were selected with G418.The cellular expression of WTX mRNA was detected using quantitative PCR and the cellular expression of WTX protein was detected by immunohistochemistry and Western blotting.Results After the mouse colorectal cancer CT26 cells were infected by lentiviral,the expression levels ofWTX mRNA and protein were significantly decreased in CT26 cells,indicating a mouse colorectal cancer CT26 cell line with stable interference of WTX gene was successfully established.Conclusion We successfully establisheda mouse colorectal cancer CT26 cell line with stable interference of WTX by lentiviral infection.关键词
