岩藻糖基转移酶2基因多态性与新疆地区哈萨克族、汉族胃腺癌发病的关系
Correlation between FUT2 gene polymorphism and risk of gastric adenocarcinoma in Kazak and Han people of Xinjiang area

Abstract
Objective To discuss the correlation between fucosyltransferase 2 (FUT2) gene polymorphism and the risk of gastric adenocarcinoma in Kazak and Han people of Xinjiang area.Methods A total of 299 cases of gastric cancer patients (case group,including 167 cases in Han and 132 cases in Kazak) and 333 healthy subjects (control group,including 182 cases in Han and 151 cases in Kazak) were included in this study.The single nucleotide polymorphisms (SNPs)of the PUT2 gene G739A,A385T and C375T loci were analyzed by PCR and DNA sequencing.We collected the clinical data,and used Multiple factors logistic regression to analyzed the relationship between the various factors and the gastric adenocarcinoma.Results In the case group and control group,there were 57 and 27 cases in Han population with AA gene at G739A locus,which were separately 32 and 18 cases in Kazak population (all P <0.01).The risk of gastric adenocarcinoma in Han and Kazak individuals with G739A locus A allele was 1.655 times and 1.430 times higher than that of the corresponding non-carriers (95% CI =1.227-2.233,P =0.001;95% CI =1.026-1.992,P =0.035).A385T locus TT genotype in Han and Kazak population were found in 7 and 12 cases,which were significantly higher than those of the same ethnic group (all P < 0.05).There was no significant difference between the Han and Kazak population of the case and control group in the T allele of C357T (all P>0.05).A385T locus T allele in Han and Kazak population of the case group were found in 39 and 45 cases (P <0.01).There was no significant difference in genotypes and allele of G739A,A385T and C357T between Kazak and Han population in the control group (all P > 0.05).Multiple regression analysis showed that the risk of gastric cancer was 2.035,2.360 and 3.815 times greater in patients with A allele at G739A locus and smoking,drinking or Hp positive.Conclusions The occurrence of gastric cancer is associated with PUT2 gene G739A,A385T polymorphism in Han and Kazak population of Xinjiang area.In patients with gastric cancer,the frequency of A385T mutation in Han is higher than that in Kazak.At the same time,smoking,drinking and Hp infection may increase the risk of gastric cancer.关键词
