Oxidized Low-Density Lipoprotein Inhibits the Expression of Notch1 and Notch2 in Human Peripheral Blood CD4+ T Cells

Abstract
Objective To explore the effect of oxidized low density lipoprotein (ox-LDL) on the expression of Notch in human peripheral blood CD4+T cells.Methods Density gradient centrifugation was used to separate the peripheral blood mononuclear cells from healthy people,and the CD4+ T cells were sorted by flow cytometry.T cells were divided into four groups:the blank control group and low,medium and high concentration group,then cocultured with ox-LDL.The concentration of ox-LDL was 0 mg/L,50 mg/L,100 mg/L and 200 mg/L,respectively.Theexpression of Notch1 and Notch2 in CD4+ T cells were detected by flow cytometry in each group after 72 h.Results The results revealed that the expression of Notch1 in each group was (3.81±0.34) %,(1.68±0.25) %,(0.73±0.21) % and (0.31±0.17) %,respectively.Compared with the blank control group,the expression of Notch1 in other groups were significantly lower,the differences had statistical significance(P<0.05).The expression of Notch2 in each group was (15.75± 1.48)%,(26.33±2.68)%,(12.42± 3.19)% and (9.87± 1.65) %,respectively.Compared with the blank control group,the expression of Notch2 was higher in the low concentration group,but lower in medium and high concentration groups,and the differences had statistical significance(P<0.05).Conclusion Ox-LDL of certain concentrations can obviously inhibit the expression of Notch1 and Notch2 in human peripheral bloodCD4+T cells,which may be the reason that ox-LDL inhibit the differentiation of Th2 and promote the differentiation of Th1,and participate in the occurrence and development of atherosclerosis.关键词
