miR-21在增生性瘢痕成纤维细胞增殖及青蒿琥酯等药物作用下表达情况的实验研究
MiR-21 in Hypertrophic Scar into the Experimental Study of the Expression of the Fibroblast Proliferation and Effect of Artesunate

Abstract
Objective To study the mechanism and of the role of miR-21artesunate inhibits human hypertrophic scar fibroblasts (HSFBs) proliferation.And provide the basis for artesunate inhibits scar hyperplasia.Methods Isolated the primary HSFBs and dealed with 24h for 100,200,300,500 μmol / L artesunate respectively.Real-time quantitative PCR (qRT-PCR) to detected miR-21 expression.Adding a certain amount of miR-21 mimics and detected the connective tissue growth factor (CTGF) and mechanism metalloproteinase 1 (MMP1) expression by qPCR and WB.Results Artesunate can inhibit the proliferation of HSFBs, and inhibit the expression of miR-21.Improve the miR-21, It significantly reduce inhibition which HSFBs dealed with artesunate by added miR-21 mimics.Meanwhile, the CTGF expression increased and the expression of MMP-1 is reduced.Conclusion Artesunate suppressed HSFBs proliferation may be regulating the expression of miR-21.关键词
