牛支原体08M株的致病性和免疫原性试验
Studies on pathogenicity and immunogenicity of Mycoplasma bovis strain 08M

Abstract
To assess the possibility of Mycoplasma bovis strain 08M as the inactivated vaccine candidate strain,pathogenicity and immunogenicity were studied.For pathogenicity test,the challenged group was intratracheally injected with 10mL M.bovis strain 08M(1× 109 CCU/mL) per calf,and the control group was intratracheally injected with 10mL0.15 mol/L pH 7.2 PBS.The nasal swabs were collected to detect the bacterium capacity using real time PCR in different time and the sera were taken to detect infection antibodies.The calves were dissected to observe the lung lesions and isolate pathogens on day 28 post-challenge.For immunogenicity test,two different antigen concentration of 0.46 and 1.48 mg/mLwas emulsified with MONTANIDE ISA 201VGadjuvant,the vaccined group was subcutaneously injected with 2mL the vaccine per calf,and the second immunization was done on day 28 after first immunization.The negative control group and the control group were injected with adjuvant and 0.15 mol/L pH 7.2 PBS,respectively.The challenge test was conducted on day 28 after the second immunization,and the method was the same as that of the pathogenicity test and the sera were collected to detect antibodies.The pathogenicity test showed that the challenged group excreted bacteria intermittently on day 4 to 14 post-challenge,infection antibodies were detected on day 7 after challenge,the lung lesions were obvious and M.bovis were isolated from them.The immunogenicity test showed that both antigen concentrations could produce high levels of antibodies after immunication,and all animals excreted bacteria intermittently after challenge.The serum antibodies reached the highest titer on day 14 post-challenge in the control group,while there were no changes in the vaccine group.Lung lesions were observed and M.bovis were isolated from all animals.In conclusion,M.bovis strain 08M has strong virulence and good immunogenicity,vaccination of calves with aninactivated vaccine made of the M.bovis strain 08M results in a strong humoral immune response but does not protect cattle from experimental challenge with M.bovis.关键词
