Cloning and Bioinformatics Analysis on CDS ofLpin2 Gene in Sheep

Abstract
In this study,the CDS region of sheep Lpin2 gene was cloned by using the RT-PCR,5'-RACE,TA cloning techniques and the bioinformatics analysis was performed,so as to provide the basic data for further exploring the genetic characters of this gene and the function mechanism of its encoding protein.As a result,the 2 754 bp length Lpin2 gene was obtained by cloning,including the 84bp length 5'UTR and the 2 670bp length CDS regionand encoding 889 amino acids.Bioinformatics analysis revealed that the encoding protein of sheep lipin2 was an unstable,neutral and hydrophilic fat-soluble proteinwith transmembrane domains,two potential O-glycosylation sites and 89 phosphorylation sites,while without signal peptide,resides in the cytosol or organelle.The potential disulphide bonds may be formed by the reactive oxygen species (ROS) produced by mitochondria in cytoplasm by the mechanism of cell redox status regulation.The secondary structure of lipin2 was mainly loop (80.54%).Two evolutionarily conserved domains of Lipin_N and LSN2 were located at the N-terminal and C-terminal of lipin2,respectively.Amino acid sequences of lipin2 were homologous between sheep and goat,cattle,alpaca,pig,dog,primates and rodents as well as chicken,and phyletic evolution was the same as their genetic relationship,which suggested that the coding region of Lpin2 gene was conservative in the course of evolution.The amino acid sequence similarity was low between lipinl and lipin2 in sheep (66%) which was possibly related to the different function activities between them.关键词
