Comparison of MPure-12 Automatic Nucleic Acid Purification and Chelex-100 Method

Abstract
Objective T o explore the forensic application value of M Pure-12 autom atic nucleic acid pu-rification (M Pure-12 M ethod) for D N A extraction by extracting and typing D N A from bloodstains and various kinds of biological sam ples w ith different D N A contents. Methods N ine types of biological sam-ples, such as bloodstains, sem en stains, and saliva w ere collected. D N A w ere extracted using M Pure-12 m ethod and C helex-100 m ethod, follow ed by PC R am plification and electrophoresis for obtaining ST R-profiles. Results T he sam ples such as hair root, chutty, butt, m uscular tissue, saliva stain, bloodstain and sem en stain w ere typed successfully by M Pure-12 m ethod. Partial alleles w ere lacked in the sam ples of saliva, and the genotyping of contact sw abs w as unsatisfactory. A dditional, all of the bloodstains (20μL , 15μL , 10μL , 5μL , 1μL ) show ed good typing results using C helex-100 m ethod. B ut the loss of alleles occurred in 1μL blood volum e by M Pure-12 m ethod. Conclusion M Pure-12 m ethod is suitable for D N A extraction of a certain concentration blood sam ples.C helex-100 m ethod m ay be better for the ex-traction of trace blood sam ples.T his instrum ent used in nucleic acid extraction has the advantages of sim plicity of operator, rapidity, high extraction efficiency, high rate of reportable ST R-profiles and low er m an-m ade pollution.关键词
