和胃安神方对失眠模型大鼠OrexinA表达的影响
Effect of Hewei Anshen Decoction(和胃安神方) on OrexinA Levels in Insomnic Rats

Abstract
Objective:To study the effect of Hewei Anshen Decoction (和胃安神方, HWASD) on orexinA levels in hypothalamus of insomnic rats.Methods:A hundred male Wistar rats were randomly divided into five groups:normal control group,insomnia model group,HWASD low dose group,HWASD middle dose group and HWASD high dose group.The content of orexinA in hypothalamus was measured by radioimmunoassav.The expression of orexinA positive neurons was detected by immunohistochemistry.Results:OrexinA levels in hypothalamus was significantly higher in insomnia model group than that in nomal group (P＜0.01).After treatment,orexinA levels was lower in HWAS low dose group than that in insomnia model group,but the difference was not statistically significant (P＞0.05).OrexinA levels were significantly lower in HWAS middle dose group and HWAS high dose group than that in insomnia model group (P＜0.01),and was not statistically different with normal control group (P＞0.05).Compared with insomnia model group,IOD of orexinA positive neurons was significantly lower in HWAS middle dose group and high dose group (P＜0.01).Conclusion:The therapeutic effect of HWASD on insomnia is partially attributed to decreased content and expression of orexinA in hypothalamus.关键词
