摘要
Abstract
Objective:To investigate the effect and mechanism of Shenqi Fuzheng injection combined with cisplatin on proliferation and apoptosis of pancreatic cancer cell line PANC-1.Methods:Cells were cultivated in vitro;PANC-1 cells were treated with Shenqi Fuzheng injection alone or combined with cisplatin.Cellular morphologic changes were observed under inverted phase contrast microscope.The effects of proliferation inhibition were assayed by MTT.The combination effect was judged through Chou-Talaly analysis.The apoptosis ratios of cells were analyzed by flow cytometry.The gene expression of Caspase-9,Fas and Survivin was detected by ELISA.Results:Shenqi Fuzheng injection alone or combined with cisplatin could inhibit the proliferation,and induce the apoptosis of PANC-1 cells.Combination of Shenqi Fuzheng injection and cisplatin revealed a synergistic effect on cell proliferation and apoptosis as it enhanced the expression of Caspase-9 and Fas but reduced that of Survivin.Conclusion:Synergistic effect of Shenqi Fuzheng injection combined with cisplatin on cell proliferation and apoptosis of PANC-l cells is possibly due to enhancement of Caspase-9,Fas and reduction of Survivin expression.关键词
