Expression of TNFR1 in human breast cancer tissue and cells and its effect on proliferation and apoptosis of MCF-7 cells

Abstract

Objective To observe the expression of tumor necrosis factor receptor-1 (TNFR1) in the human breast cancer tissues and cells and the effect of inhibiting TNFR1 expression on the proliferation and apoptosis of MCF-7 cells, and to discuss the mechanism.Methods The TNFR1 mRNA expression was detected in 30 cases of human breast cancer tissues and adjacent tissues, and cells (MCF-7, MDA-MB-231 and T47D) by real-time PCR.MCF-7 cells were divided into 3 groups: groups A, B and C.Group A (control group) was not transfected, group B was transfected with negative vector and group C was transfected with siRNA-TNFR1 vector.After transfection for 72 h, MTT and flow cytometry were used to detect proliferation and apoptosis of MCF-7 cells, respectively;and Western blot was performed to determine the Caspase-8 and nuclear factor-κB (NF-κB).Results The relative expression of TNFR1 in the human breast cancer tissues and adjacent tissues was 1.41±0.08 and 0.38±0.05, respectively.The relative expression of TNFR1 in MCF-7, MDA-MB-231, T47D and normal breast cells was 2.48±0.06, 1.51±0.02, 1.66±0.01 and 1.00±0.09, respectively.TNFR1 expression in the breast cancer tissues and cells was higher than that of the control group (P<0.05).The optimal density in proliferation of groups A, B and C was 0.96±0.09, 0.93±0.11 and 0.62±0.05, respectively.The apoptosis rates of groups A, B and C were 8.23%±3.50%, 9.92%±3.19% and 24.61%±2.46%, respectively.Significant difference was found in the proliferation and apoptosis rate between groups A, B and C (all P<0.05).The Caspase-8 expression in the groups A, B and C was 0.40±0.03, 0.42±0.05, 0.82±0.01, and the NF-κB protein expression was 0.79±0.03, 0.76±0.04 and 0.32±0.06, respectively.Significant difference was found between groups A, B and C (all P<0.05).Conclusion TNFR1 is highly expressed in human breast cancer tissues and cells and probably promotes the cell proliferation and suppresses the apoptosis by activating NF-κB signaling pathway and inhibiting Caspase-8 expression.
