拟态弧菌溶血素VMH蛋白B细胞线性表位的预测与鉴定
Prediction and identification of linear B-cell epitopes of Vibrio mimicus hemolysin VMH protein

Abstract
In order to locate the B-cell linear epitopes of Vibrio mimicus heat-labile hemolysin protein (VMH),B-cell linear epitope prediction was performed with comprehensive ABCpred and BepiPred web servers according to the full-length amino acid sequence of the protein.Meanwhile,taking cytolysin protein from Vibrio.cholerae as a template,a 3D structure model of VMH protein was constructed using Modeller 9.14 software,and the potential B-cell epitopes were represented for determination of their position in the 3D structure using PyMol software.The results showed that the six most likely epitopes,which were located in N-terminal regions of P130-141,P192-203,P236-247,P281-292,P425-436,P464-475,respectively and entirely or mostly exposed on the surface of the 3D structure model of VMH protein,were obtained after consideration of multiple factors.The six predicted epitope peptides were artificially synthesized and identified by peptide ELISA,inhibitory Western blotting and hemolytic activity assay.It was found that the six predicted epitope peptides could react to the rabbit anti-VMH protein antibody.Except predicted epitope P281-292,other five predicted epitopes could significantly inhibit the immunoreaction between anti-VMH antibody and VMH protein in Western blotting and have hemolysis activity.These results indicated that predicted epitope P130-141,P192-203,P236-247,P425-436 and P464-475 were indeed the linear B-cell epitopes of VMH protein from V.mimicus.关键词
