PI3K抑制剂LY294002联合吡柔比星对膀胱癌BIU-87细胞增殖作用的影响
Effects of the proliferation of human bladder cancer cells BIU-87 after PI3K inhibitor LY294002 and pirarubicin combined treatment

Abstract
Objective:To investigate the effects of PI3K inhibitor LY294002 combined with Pirarubicin (THP) on proliferation of human bladder cancer cells BIU-87.Methods:The BIU-87 cells were treated by the different concentration of LY294002 and THP for 0,12,24,36 h.The tumor inhibition rate was measured with MTT.The Bax,Bcl-2,Caspase-3 and P-gp of BIU-87 cells were detected by Western blotting.Results:THP could significantly inhibit the proliferation of BIU-87 cells and had a significant time dependence (P<0.05).Compared the THP combined with different concentration of LY294002 groups and used alone at the same dose in THP group,the proliferation of BIU-87 cells increased with the increase of drug concentration and time further inhibited,and the low concentration THP group combined with LY294002 on the inhibition of BIU-87 cells was stronger than the high dose THP group (P<0.05).Furthermore,the expressions of Bax and Caspae-3 in cytoplasm of BIU-87 cells treated by LY294002 and THP were increasing more than the control group(P<0.05),while the expressions of Bcl-2 and P-gp were decreasing(P<0.05).Conclusion:It is better inhibiting effect on BIU-87 cell to use LY294002 and THP combined treatment than only THP.关键词
