摘要

Abstract

Aim: To investigate the protection effect of phyllaemblicin B on myocardiocytes and Balb/c mice infected with coxsackie virus B3(CVB3).Methods: Viral myocarditis(VMC) models infected with CVB3 were established in myocardiocytes and Balb/c mice.Cell survival rate was measured by cytopathic effect(CPE) assay, the contents of aspartate transferase(AST) and lactate dehydrogenase(LDH) were determined in supernatant of myocardiocytes culture medium.Histopathological changes of myocardium of Balb/c mice infected with CVB3 were analyzed to test the antiviral activity of phyllaemblicin B.Results: CVB3 infection induced obvious CPE in myocardiocytes in vitro.After treatment with phyllaemblicin B, CPE on myocardiocytes infected with CVB3 was obviously inhibited, meanwhile the activities of AST and LDH were dramatically reduced compare with the untreatment group(P<0.05).Ultra-structure of mouse myocardium demonstrated that CVB3 infection led to pathological lesions including cardiac muscle fibers breakdown and destruction of nucleus structure.With the treatment of phyllaemblicin B, the histopathology changes in mice myocardium were alleviated.Conclusion: Phyllaemblicin B has obvious protection effect on myocardiocytes and Balb/c mice infected with CVB3.
