Identification of Differentially Expressed Proteins in Lung Adenocarcinoma by Laser Capture Microdissection and Proteomic Analysis

Abstract
To screen for novel proteins associated with the pathogenesis of lung adenocarcinoma (AdC),laser capture microdissection (LCM) was applied to capture and collect target cells from AdC and normal bronchial epithelium (NBE) tissues,respectively.Two-dimensional gel electrophoresis (2-DE) was performed to separate proteins of microdissected AdC and NBE.PDQuest software system was used to analyze 2-DE images,and differential protein spots between the 2 types of cells were analyzed by matrix-assisted laser desorption/ionization-time of flight-mass spectrometry (MALDI-TOF-MS).Immunohistochemistry of tissue microarray was conducted to detect the expression of the differential protein annexin A4 in 30 cases of AdC tissues,their paracancerous lung (PCL) tissues and matched positive node metastasis (PNM) tissues.The results showed that 2-DE patterns of microdissected AdC and NBE were established by proteomic technology,and 33 differential proteins between AdC and NBE were identified,21 of which were up-regulated and 12 of which were down-regulated in AdC.What's more,immunohistochemistry showed that annexin A4 was significantly overexpressed in the AdC tissues compared with their PCL tissues,and that annexin A4 expression in matched lymph node metastases were higher than the primary tumour.The data suggest that annexin A4 might play an important role in AdC progression,and serve as a novel biomarker for lymph node metastasis in AdC.关键词
