Expression of Fat-1 gene inhibits the growth of human endometrial cancer Ishikawa cells in vivo

Abstract
Objective:To explore the inhibiting effect of adenovirus mediated Fat-1(Ad-GFP-Fat-1)gene transfer in human endometrial carcinoma Ishikawa cells and the potential molecular mechanism in vivo.Methods:Human endometrial cancer Ishikawa cells were implanted subcutaneously into the right armpit of each nude mouse.The models of xenografted tumor were established through the subcutaneous inoculation of nude mice,which were then randomly divided into three groups:PBS control group,Ad-GFP empty vector group and Ad-GFP-Fat-1 experiment group.Mice in all groups were intratumorally injected and the anti-tumor efficacy was evaluated by measuring tumor volume and weight.Then RT-PCR was used to analyze Fat-1 gene expression in tumors.The apoptosis of cells in xenografted tumor tissues was detected by TUNEL+DAPI staining method.The expression of cysteineprotease (Caspase)-3 was detected by immunohistochemical SP method and Western blot.Results:Fat-1 gene was successfully transfected and expressed in the tumor tissues of Ad-GFP-Fat-1 group.Compared with the other two groups,the tumor volume and weight of Ad-GFP-Fat-1 group were reduced(P<0.05 for both).The apoptosis index in Ad-GFP-Fat-1 group was obviously higher than that in PBS group and Ad-GFP group (P<0.05).The Ad-GFP-Fat-1 can obviously increase the expression of Caspase-3.Ad-GFP-Fat-1 increased the expression of Caspase-3 mainly by increasing the activation of Caspase-3 (Cleaved Caspase-3).Conclusion:The expression of Ad-GFP-Fat-1 can inhibit the growth of subcutaneous human endometrial cancer xenografts in nude mice,its mechanism may be related to Fat-1 gene raised Caspase-3 expression induced apoptosis.关键词
