The development of pinopodes and expression of integrin beta 3 in the endometrium of rat model of intrauterine adhesion

Abstract
Objective:To establish a rat model of intrauterine adhesions,observe the development of pinopodes and the expression of integrin beta 3 in the endometrium.To investigate the effect of intrauterine adhesion on endometrial receptivity.Methods:26 healthy SD rats were selected,and each rat left uterus as the model group (injected with absolute alcohol and continued 5minutes),the right uterus as control group (injected with normal saline).The uterus was removed after two estrogen cycles.The change of endometrial morphology and the degree of endometrial fibrosis was observed.The development of pinopodes in endometrium was observed by scanning electron microscope and the expression of integrin beta 3 was detected by immunohistochemical method.Results:In the model group,the endometrium was obviously thinner,and the epithelial cells arranged in disorder,and the collagen fibers in the stroma increased.The endometrial fibrosis score was 4.69±0.22 points,significantly higher than the control group.By scanning electron microscope,we found there were a large number of fully developed pinopodes in the endometrium of the control group,while the endometrial pinopode in the model group sparse distribution,surface collapse,and developed non-synchronized.The expression of integrin beta 3 in the model group was significantly lower than that in the control group,and the difference was statistically significant (P<0.05).Conclusion:A stable animal model of intrauterine adhesion can be established by injecting absolute alcohol,which inhibit the expression of endometrial pinopode and integrin beta 3,and reduce the endometrial receptivity.关键词
