电针联合身痛逐瘀胶囊对CCI模型大鼠疼痛及脊髓组织GFAP表达的影响
Effect of Electroacupuncture Combined with Shentong Zhuyu Capsule on Pain and Expression of GFAP in CCI Model Rats

Abstract
Objective:To observe the effect of electroacupuncture combined with Shentong Zhuyu capsule on the threshold of mechanical compression and the expression of GFAP in spinal cord tissue of CCI rats in order to reveal its mechanism from the inhibition of glial cell activation in spinal cord tissue.Methods:Adult SD rats were randomly divided into sham operation group,model group,electroacupuncture group,traditional Chinese medicine group and acupuncture combined with Chinese medicine group.Each group was divided into three subgroups:3 days,7 days and 14 days,with 10 rats in each subgroup.According to Bennett method to establish CCI rat models,electroacupuncture group received Sunshang Cehuan Tiao therapy,Yanglingquan electroacupuncture treatment,once per day with 30min.Chinese medicine group received Shentong Zhuyu capsule treatment (1.64 g/kg),administered by intragastric administration,administration volume of 15 ml/kg,once per day.Acupuncture combined with Chinese medicine group received acupuncture and traditional Chinese medicine treatment,with the same methods and treatment above.Sham operation group and model group were given equal volume of saline.The expression of GFAP mRNA in the spinal cord of CCI model rats was detected by RT-PCR.The expression of GFAP mRNA was detected by RT-PCR.Results:The MWT test showed that the MWT of the model group was significantly decreased after operation of 3 days,and there was a significant difference between two groups (P < 0.05).In the acupuncture group,the traditional Chinese of medicine group,the acupuncture combined with Chinese medicine group MWT of CCI rats was increased,and the difference was statistically significant (P < 0.05) at each time point compared with that in the model group.The MWT of acupuncture combined with Chinese medicine group was significantly higher than that of the acupuncture group and the traditional Chinese medicine group (P < 0.05).The relative expression of GFAP mRNA in the spinal cord tissue was significantly higher than that in the sham operation group (P < 0.05),and the expression of GFAP mRNA in the model group was significantly higher than that in the sham operation group (P < 0.05).The relative expression of GFAP mRNA in spinal cord tissue of acupuncture group,Chinese medicine group and acupuncture combined with Chinese medicine group was significantly lower than that of model group (P < 0.05).The relative expression of GFAP mRNA in the acupuncture combined with Chinese medicine group was significantly lower than that in the acupuncture group and the traditional Chinese medicine group (P < 0.05).The results showed that acupuncture group,traditional Chinese medicine group and acupuncture combined with Chinese medicine group could inhibit the expression of GFAP mRNA in spinal cord tissue of CCI model rats,especially in acupuncture combined with Chinese medicine group.Conclusion:Electroacupuncture combined with Shentong Zhuyu capsule can significantly improve the threshold of mechanical compression in CCI model rats,and its mechanism may be related to the inhibition of GFAP mRNA expression in spinal cord tissue.关键词
