Abstract

This experiment was conducted to investigate the effect of dietary iron (Fe) level on manganese (Mn) absorption and deposition in broilers.A total of 336 1-day-old Rose-308 male chicks were divided into 4 groups with 6 replicates and 14 chicks each replicate cage,and fed the basal diet (control,containing Fe 77.7 mg/kg) or basal diet supplemented with 100,250,or 500 mg Fe/kg for 28 days.Ligated duodenal loops were perfused with 8.74 mmol/L of Mn to determine the Mn absorption.The results showed that plasma Fe content and transferrin saturation increased (P<0.10) as dietary Fe level increased.The Fe contents in pancreas,duodenal mucosa,and tibia ash increased (P<0.10) and the Mn contents in above mentioned tissues decreased (P<0.10) as dietary Fe level increased.The DMT 1 mRNA levels of Fe-supplemented groups were lower (P<0.10) than that of the control,and the FPN1 mRNA level of Fe-supplemented 500 mg/kg group was lower (P<0.10) than those of the control and the other two Fe-supplemented groups.The Mn absorption rates of Fe-supplemented groups were lower (P<0.10) than that of the control.These results suggested that dietary Fe might decrease the absorption and deposition of Mn through the down-regulations of DMT1 and FPN1 gene expression in duodenal mucosa of broilers.
