摘要

Abstract

Objective:To study the expression of p62 in nasopharyngeal carcinoma (NPC) cell lines and tissues,and explore the serum concentration of anti-p62 in NPC patients,and investigate the correlation between p62 and NPC occurrence and metastasis,and assess its clinical value.Methods:The expression of protein p62 in NPC cell lines (NP69,6-10B and 5-8F) was detected by Western blot.Immunohistochemistry staining method was used to test the expression of p62 in NPC tissue specimens.Serum anti-p62 antibody level was determined by enzyme-linked immunosorbent assay,then analyzing its relationship with clinical pathological features.Results:The expression of p62 in NP69,6-10B and 5-8F cells was increased successively (P<0.05);the expression of p62 was increased in nasopharyngeal carcinoma tissues,which was closely correlated with metastasis (x2=9.332,P=0.002).Compared with chronic rhinopharyngitis group,the serum level of anti-p62 in NPC patients was dramatically increased (t=-4.653,P<0.001) and correlated with metastasis (t=14.255,P=0.016);the optimal value of serum anti-p62 screening for NPC is 51.82 μg/L,and the critical value of anti-p62 judging NPC patients whether metastasis or not is 62.03 μg/L.Conclusion:High expression of p62 is closely related to the occurrence and metastasis of NPC,p62 protein in tissues and serum anti-p62 may be used as new indicators for monitoring the conditions and prognosis of NPC patients.
