Effects of CYB5R2 on apoptosis, cell cycle and invasion of nasopharyngeal carcinoma cells

Abstract
Objective:To explore the influences of CYB5R2 gene on cell apoptosis,cell cycle and invasion and metastasis ability of nasopharyngeal carcinoma (NPC) cells.Methods:CYB5R2-pCMV-Tag3A and corresponding empty vector were transfected into CNE2 and HONE1 cells.The expression of CYB5R2 was confirmed by RT-PCR.Cell apoptosis and cycle were detected by flow cytometry.The activity of Caspase 8 and Caspase 9 were detected by colorimetric assay.Trans well invasive assay was used to evaluate the invasiveness of NPC cells.Results:The activity of Caspase 8,Caspase 9 and apoptosis rate in CYB5R2 overexpressing cells were significantly higher than those in control groups (P＜0.05).Compared with control,the proportion of S phase deduced while the proportion of G0/G1 phase was significantly increased in the CYB5R2 overexpressing cells.The number of invading cells in CYB5R2 overexpression group was significantly decreased as well (P＜0.05).Conclusion:Overexpression of CYB5R2 promotes cell apoptosis,arrests cell cycle and suppresses invasiveness of NPC cells,suggesting that CYB5R2 could be a potential antioncogene in treating NPC.关键词
