芍药苷通过TLR4炎症通路和Nrf2氧化应激通路减轻大鼠脊髓损伤的实验研究
Experimental study on reducing spinal cord injury in rats by TLR4 inflammatory pathway and Nrf2 oxidative stress pathway

Abstract
Objective: To study the effecof paeoniflorin on TLR4 inflammatory pathway and Nrf2 oxidative strespathway in ramodel with spinal cord injury.Methods: Male SD ratwere selected and divided into sham group, spinal cord injury group and paeoniflorin group, spinal cord injury ramodelwere established and then given 30 mg/kg paeoniflorin fo14 d of intervention in row, and then serum and spinal cord specimenwere collected to determine the expression of cell apoptosigenes, TLR4 pathway geneand Nrf-2 pathway genes.Results: Spinal cord caspase-3, caspase-8, caspase-9, TLR4, MyD88, NF-kB, AP-1, Nrf-2 and ARE mRNexpression awell aserum IL-6, TNF-α and IL-12 contentof spinal cord injury group were significantly highethan those of sham group while spinal cord HO-1, SOD and GSH-PX contentwere significantly lowethan those of sham group;spinal cord caspase-3, caspase-8, caspase-9, TLR4, MyD88, NF-kand AP-1 mRNexpression awell aserum IL-6, TNF-α and IL-12 contentof paeoniflorin group were significantly lowethan those of spinal cord injury group while spinal cord Nrf-2 and ARE mRNexpression awell aHO-1, SOD and GSH-PX contentwere significantly highethan those of spinal cord injury group.Conclusions: Paeoniflorin can inhibithe TLR4 inflammatory pathway and enhance the Nrf2 anti-oxidative strespathway to reduce the spinal cord injury and inhibicell apoptosiin rats.关键词
