摘要

Abstract

Objective: To study the effecof sunitinion renal cancecell growth in vitro and invasion awell aWnt/β-catenin signaling pathway.Methods: Renal cancecell lineACHN were cultured and processed with differendoseof sunitini(1 μmol/L, 2 μmol/L, 4 μmol/L and 8 μmol/L), and sunitinib-free processing condition waused anegative control.Twenty-fouhourafteprocessing, the mRNexpression levelof apoptosigenes, invasion geneand Wnt/β-catenin signaling pathwayin cellwere detected.Results: Twenty-fouhouraftetreatment, NPRL2, Bax, caspase-3 and caspase-9 mRNexpression in 1 μmol/L, 2 μmol/L, 4 μmol/L and 8 μmol/L sunitinigroupwere significantly highethan those in negative control group while MMP2, MMP9, Vimentin, N-cadherin, Wnand β-catenin mRNexpression were significantly lowethan those in negative control group;the highethe dose of sunitinib, the highethe NPRL2, Bax, caspase-3 and caspase-9 mRNexpression while the lowethe MMP2, MMP9, Vimentin, N-cadherin, Wnand β-catenin mRNexpression in cells.Conclusions: Sunitinican inhibithe Wnt/β-catenin signaling pathway in renal cancecellto increase the expression of apoptosigenes, inhibithe expression of invasion geneand thereby inhibithe cell growth and invasion.
