Expression and Purification of Protein all3556 from Anabaena sp.PCC 7120

Abstract
Succinic semialdehyde dehydrogenase plays important physiological roles in the prokaryotes and eukaryotes.BLAST sequence analysis reveals that the gene all3556 in Anabaena sp.PCC 7120 encodes succinic semialdehyde dehydrogenase.The gene all3556 was cloned from the genome of Anabaena sp.PCC 7120 by conventional PCR.Then the plasmid of pET-28a-all3556 was constructed by ligating all3556 gene with pET-28a vector, and the expression of all3556 in the competent cells of Escherichia coli strain BL21 (DE3) was observed.The results of SDS-PAGE showed that the protein all3556 could high-efficiently express in soluble form in this system.The protein all3556 was purified by Ni-NTA affinity chromatography, its purity was more than 95%, and its yield was about 27 mg/g wet cell.This study will lay an important base for further illustrating the catalytic function and mechanism of all3556 protein in the future.关键词
