花生枝叶水煎液对大鼠脑缺血再灌注模型的保护作用及机制研究
Study on protective effect and mechanism of decoction of branches and leaves of Arachis hypogaea L on model of cerebral ischemia-reperfusion injury in rats

Abstract
Objective To investigate the potential neuroprotective effect and mechanism of decoction of the branches and leaves of A rachis hypogaea L on model of cerebral ischemia-reperfusion injury (CI-RI) in rats.Methods Rats were grouped as follows:control,sham operation,model,and low,moderate and high dose [(50,100 and 150 g/(kg· d),respectively] of decoction of the branches and leaves ofArachis hypogaea L.CI-RI model was established by modified suture occlusion of middle cerebral artery (MCAO).The neuroprotective effect of decoction of the branches and leaves of Arachis hypogaea L was evaluated by the neurological deficit score,cerebral infarct volume,and levels of malondialdehyde (MDA),reactive oxygen species (ROS),glutathione peroxidase (GPx),superoxide dismutase (SOD) in brain tissue.Results Compared with the model group,groups with moderate and high doses of decoction of the branches and leaves of Arachis hypogaea L could reduce both the neurological deficit score [(2.27±0.80),(1.93±0.80) and(1.73±0.80),respectively] and cerebral infarct volume [(42.46%±6.31%),(32.20%±7.87%) and (24.80%±4.91%),respectively],and decrease the levels of MDA [(519.66±54.31),(445.87±12.05) and (303.98±36.03) nmol/mg protein,respectively] and ROS [(2.84±0.12),(2.10±0.10) and (1.37±0.04) fluorescence intensity/mg protein,respectively],increase the activities of GPx [(32.47± 1.92),(66.50±6.04) and (86.39±6.35) U/mg protein,respectively)] and SOD [(72.53±11.27),(165.14±14.05) and (258.62±39.21) U/mg protein,respectively] in brain tissue.Conclusions Decoction of the branches and leaves of Arachis hypogaea L has potential neuroprotective effect on model of CI-RI in rats,and the mechanism is correlated with its antioxidative stress effect.关键词
