摘要

Abstract

Objective To detect the combining of hemoglobin with arsenic and analyze the effect of hemoglobin on inhibitory effect of arsenic trioxide (ATO) on platelet aggregation.Methods The combining of hemoglobin with paminophenylarsine oxide (PAPAO) was detected with electrospray ionization mass spectrometry (ESI-MS).Whole blood or platelet rich plasma (PRP) was incubated with ATO (0,10 μmol/L).Different concentrations of hemoglobin (0,40,60,80,100,120 g/L) were added into PRP,and incubated with ATO (0,10 μ mol/L).Then the platelet aggregation induced by adenosine diphosphate (ADP) (2 μmol/L) or collagen (2 mg/L) was measured.Results β chain of hemoglobin could combine with one molecule of PAPAO via a dehydration reaction.ATO could inhibit platelet aggregation in both PRP and whole blood,the inhibitory effect was significantly higher in PRP.Hemoglobin (≥ 80 g/L) could concentration dependently reverse the inhibition of platelet aggregation induced by collagen(r=0.956,P=0.003).Similarly,hemoglobin (≥ 80 g/L) could concentration dependently reverse the inhibition of platelet aggregation induced by ADP (r=0.940,P=0.005).Conclusions Hemoglobin reduces the inhibitory effect of ATO on platelet aggregation via binding to arsenic in a concentration-dependent manner.Hemoglobin plays an important role in the process of ATO inhibition of platelet aggregation.
