Sodium Houttuyfonate Induces Apoptosis of Candida albicans Biofilm Cells with Clinical Drug Resistance

Abstract
Objective To investigate the effect of sodium houttuyfonate on the apoptosis of Candida albicans biofilm cells with clinical drug-resistance.Methods DAPI staining were performed to observe the morphology of drug-resistant Candida albicans biofilm cell nuclei.FITC-VAD-FMK staining assay was performed to evaluate metacaspase activity in drug-resistant Candida albicans biofilm cells.With the help of JC-1 and DCFH-DA staining, flow cytometry was used to measure mitochondrial membrane potential (MMP) and reactive oxygen species (ROS) level in drug-resistant Candida albicans biofilm cells.Annexin Ⅴ-FITC/PI staining and flow cytometry were used to observe the apoptosis rate of drug-resistant Candida albicans biofilm cells.Results Under the action of sodium houttuyfonate at a certain concentration, drug-resistant Candida albicans biofilm cells had significant increases in ROS level, cell apoptosis rate, and metacaspase activity and a significant reduction in MMP, with fragmentation and shrinkage of cell nucleus.Conclusion Sodium houttuyfonate can induce the apoptosis of drug-resistant Candida albicans biofilm cells.关键词
