摘要

Abstract

Objective To investigate the effect of sodium houttuyfonate on the apoptosis of Candida albicans biofilm cells with clinical drug-resistance.Methods DAPI staining were performed to observe the morphology of drug-resistant Candida albicans biofilm cell nuclei.FITC-VAD-FMK staining assay was performed to evaluate metacaspase activity in drug-resistant Candida albicans biofilm cells.With the help of JC-1 and DCFH-DA staining, flow cytometry was used to measure mitochondrial membrane potential (MMP) and reactive oxygen species (ROS) level in drug-resistant Candida albicans biofilm cells.Annexin Ⅴ-FITC/PI staining and flow cytometry were used to observe the apoptosis rate of drug-resistant Candida albicans biofilm cells.Results Under the action of sodium houttuyfonate at a certain concentration, drug-resistant Candida albicans biofilm cells had significant increases in ROS level, cell apoptosis rate, and metacaspase activity and a significant reduction in MMP, with fragmentation and shrinkage of cell nucleus.Conclusion Sodium houttuyfonate can induce the apoptosis of drug-resistant Candida albicans biofilm cells.
