金线莲乙醇提取物抗氧化作用的研究
Study on antioxidant activity of ethanol extract from Anoectochilus Formosanus

Abstract
Objective To study the effect of ethanol extract of Anoectochilus Formosanus (AF) on antioxidation in natural aging mice and its antioxidant activities in vitro.Methods Eight month-old KM mice were randomly divided into 5 groups,including the natural aging group,the positive control group (vitamin E group,0.1 g/kg) and three different doses of AF ethanol extract groups (1,2 and 4 g/kg).The animals in each group were given the corresponding dose of drugs daily,and the natural aging group was given the same amount of normal saline for 30 days.The body weight of mice was determined before administration and after administration of 14 d and 28 d.The levels of MDA,GSH-PX and SOD in serum and liver were measured.The total antioxidant capacity,hydroxyl radical (·OH),superoxide anion (O-·2) reaction system were used to study the antioxidant activity of AF ethanol extract.Results Compared with the natural aging group,there was no significant difference in body weight in AF ethanol extract-treated groups (P>0.05).The activities of blood GSH-PX and T-SOD were significantly increased in vitamin E and AF ethanol extract groups (P<0.01 or P<0.05).The levels of MDA were decreased in vitamin E and high and middle doses of AF ethanol extract groups (P<0.01 or P<0.05).The activities of liver GSH-PX and SOD,but not MDA,were significantly increased in vitamin E and high dose of AF ethanol extract groups (P<0.01).The activities of liver SOD were significantly increased and MDA content was decreased in the middle dose of AF ethanol extract group.The effects of AF ethanol extract in scavenging hydroxyl free radical,superoxide anion and total antioxidant capacity were dose-effect relationship in a certain concentration range.In hydroxyl and superoxide anion assays,the IC50 of AF ethanol extract were 5.5 mg/mL and 12.5 mg/mL.Conclusion Ethanol extract of AF displays an antioxidation effect.关键词
