Effects of Cyclohexanhexol-derived Synthetic Ice Blocker and Trehalose on Frozen Goat Sperm

Abstract
In order to reduce the physical and chemical damages on goat sperm during freezing and thawing,the protective effects of cyclohexanhexol-derived synthetic ice blocker (SIB) and trehalose on frozen goat sperm were evaluated and compared in this study.The used SIB was 1,4-cyclohexanediol (1,4-CHD).Semen was collected from six Yunnan Black goats using an artificial vagina.After repeated washing by centrifugation to remove seminal plasma,the cell pellet was mixed with the freezing extender.The sperm was directly plunged into liquid nitrogen after equilibration at 4℃ and prefreezing in liquid nitrogen vapor.The post-thaw sperm motility,acrosome,plasma membrane,and tolerance to hypotonicity were determined.At the same time,the transmission electron microscope (TEM) was used to observe the ultrastructural alterations of frozen-thawed sperm.The data indicated that as compared to the control group(35.97%±6.21%,55.46%±8.91%,and 34.17%±5.68%),1,4-CHD and trehalose could significantly improve the post-thaw motility(41.35%±8.14%,43.59%±6.23%),acrosome integrity(62.73%±7.62%,64.29%±7.81%) and tolerance to hypotonicity (40.29%±7.41%,39.79%±6.58%)of goat sperm (P<0.05).Moreover,trehalose could inhibit cell apoptosis caused by freezing and thawing.However,the presence of 1,4-CHD could not decrease the cryo-injuries on the membrane integrity and the phosphatidylserine (PS) distribution of goat sperm.As compared to 1,4-CHD,trehalose could significantly reduce the damages caused by freezing and thawing on sperm membrane.The TEM results indicated that freezing could seriously injure sperm membrane and acrosome.The post-thaw sperm membrane was swollen,lost or broken.The freezing and thawing process can result in rupture or loss of outer acrosome membrane.In addition,partly or complete loss of acrosomal contents had occurred in some post-thaw spermatozoa.The TEM results further proved that the protective effect of trehalose on frozen sperm membrane was superior to that of 1,4-CHD.In conclusion,this study further proved that the cyclohexanhexol-derived SIB and trehalose could reduce the cryo-injuries on goat sperm.However,as compared to trehalose,cyclohexanhexol-derived SIB,such as 1,4-CHD,could not mitigate the damages on sperm membrane during freezing and thawing.Therefore,how to reduce the negative effects of freezing and thawing on goat sperm membrane still needs further research.关键词
