摘要
Abstract
Objective To investigate the changes of chromosome 8 (8p) in primary hepatocellular carcinoma(HCC) and its metastatic foci to provide a guidance for the screening of HCC metastasis related gene targets.Methods The oligonucleotide microarray containing all the genes on chromosome 8p was prepared and mixed RNA in 10 cases of normal liver tissue to form a sample pool as the control sample for the detection of 10 cases of non-metastatic HCC tissues and 10 cases of metastatic lesions.The expression of the related genes in HCC tissues was further verified in the cell lines with different metastatic potentials,and the corresponding genes on chromosome 8P and metastatic lesions of liver cancer were analyzed by the hybridization system of chip hybridization.The reverse transcription polymerase chain reaction(RT-PCR) and real-time PCR were used to validate the selected genes.Results Compared with the normal liver tissue,the chromosome 8p gene expression in different metastatic potentials of HCC tissue had difference,Fgli,Clu,Msra,Defbi and other genes were significantly down-regulated in HCC metastatic lesions when the fluorescence intensity was set to >800,RT-PCR further indicated that they were down-regulated in HCC metastatic lesions.Conclusion Chromosome 8p is associated with metastatic foci of HCC.Fgli,Clu,Msra and Defbi loci can be used as candidate genes to inhibit the metastasis of HCC,which is worthy of clinical deep study.关键词
