低聚原花青素对大鼠肝纤维化氧化损伤的影响
Effects of oligomeric proantho cyanidins on oxidative damage of rat liver fibrosis

Abstract
Objective To study the effect and mechanism of oligomeric proantho cyanidins on hepatic fibrosis.Methods SD rats were divided randomly into 6 groups:control group,model group,Fuzheng Huayu Capsule group,oligomeric proantho cyanidins group(60,120,240 mg·kg-1).The hepatic fibrosis model was established by intraperitoneal injection of carbon tetrachloride(CCl4).Liver pathology was observed by hematoxylin-eosin (HE) staining.Biochemical method decect Serum alanine aminotransferase(ALT) and aspartate transferase(AST).The contents of superoxide dismutase(SOD) and malondialdehyde(MDA) in liver tissue were determined by enzyme-linked immunosorbent assay(ELISA).Western blot assay was used to detect Sirt1 and Nrf2 protein expression.Results Oligomeric proantho cyanidins(60,120,240 mg·kg-1)significantly inhibited CCl4-induced liver fibrosis in rats.The ALT and AST level in serum,MDA level in liver were significantly reduced,SOD significantly increased.Furthormore,Oligomeric proantho cyanidins were upregulated Sirt1 and Nrf2 protein expression.Conclusion Oligomeric proantho cyanidins have anti-fibrotic effects,its mechanism maybe related to its antioxidant effects.关键词
