摘要

Abstract

Objective To observe the expression of RASSF10 gene in renal cell carcinoma (RCC) and its promoter methylation, and to investigate its significance. Methods The mRNA expression and methylation rate of RASSF10 gene in RCC tissues, para-carcinoma tissues, and normal tissues of 52 cases were detected by using reverse transcription polymerase chain reaction (RT-PCR) and methylation-specific polymerase chain reaction (MSP). Their relationship with pathological features of RCC patients and the relationship between mRNA expression and methylation of RASSF10 gene were analyzed. Results The mRNA relative expression level of RASSF10 gene in RCC tissues was lower, and the methylation rate was higher than that in the para-carcinoma tissues and normal tissues (both P<0.05). The mRNA expression and promoter methylation rate of RASSF10 gene in RCC tissues were not correlated with age, histological type, tumor size, and Fuhrman grade (all P>0.05), but were correlated with tumor TNM grade (P<0.05). The mRNA relative expression of RASSF10 gene in RCC patients with methylation was lower than that without methylation (P<0.05).Conclusion The low expression of RASSF10 gene in RCC tissues may be related to promoter hypermethylation, and low expression of RASSF10 gene and its promoter methylation may participate in the occurrence and development of RCC.
