miR-150过表达对小鼠垂体瘤细胞株GT1-1增殖的调控作用及机制
Regulation of overexpression of miR-150 on proliferation of GT1-1 cell line in mice with pituitary adenoma

Abstract
Objective To investigate the regulation of overexpression of miR-150 on proliferation of GT1-1 cell line in mice with pituitary adenoma and its mechanism.Methods GT1-1 cells were divided into the overexpression group and the negative control group, which were then transfected with miR-150 mimics and miR-150 mimics NC for 48 hours, respectively. The proliferation was detected with MTT, the relative expression of the thrombospondin 1 (TSP1) and transforming growth factor β1 (TGF-β1) proteins in GT1-1 cells was detected by Western blotting, the expression of latent TGF-β1 and active TGF-β1 in the supernatant was detected by ELISA, and the target gene of miR-150 was detected with the Dual-Luciferase reporter gene system.Results After 48-hour transfection, the proliferation rate in the overexpression group was significantly lower than that of the negative control group (P<0.05). The relative expression of the TSP1 and TGF-β1 proteins in the overexpression group was significantly lower than that of the negative control group (P<0.05). Significant difference was found in the expression of latent TGF-β1 and active TGF-β1 in the supernatant of cultured cells between the overexpression group and the negative control group (both P<0.05). The miR-150 bound with 3′-UTR of TSP1 mRNA, and TSP1 was the target gene of miR-150.Conclusion The overexpression of miRNA-150 may inhibit the proliferation of GT1-1 cells mediating its target TSP1/TGF-β1 pathway.关键词
