加味四妙散汤剂对大鼠急性痛风性关节炎的疗效及可能作用机制的研究
Effect and potential mechanism of modified Simiao decoction in rat models with acute gouty arthritis

Abstract
Objective To evaluate the effect and potential mechanism of modified Simiao decoction (MSD) in rat models with acute gouty arthritis.Methods The rat models with acute gouty arthritis were established and divided into the model,colchicin (PC),low-dose (MSD1) and high-dosage MSD (MSD2) and blank control groups(BC).Rats in different groups were given with corresponding medicine via intra-gastric approach.The index of joint swelling in the right ankle was measured at 0,6,24,48 and 72 h.At 3 d after medicine administration,the synovial fluid was collected for the detection of IL-1β,IL-6 and TNF-α levels by ELISA.The rats were sacrificed and the soft tissues surrounding the joint were sampled and subject to histological examination of the infiltration of inflammatory cells.Results At different time points,the index of joint swelling in the model group was significantly higher compared with that in the BC group (all P<0.05).Compared with the BC group,joint swelling was observed at 6 h and peaked at 24 h after MSD administration in the model group.At 24 h following MSD administration,the index of joint swelling in the PC,MSD1 and MSD2 groups was significantly lower than that in the model group (all P<0.05).The index of joint swelling in the MSD1 and MSD2 groups was considerably higher compared with that in the PC group (all P<0.05).The index of joint swelling in the MSD2 group was significantly lower than that in the MSD1 group at different time points (all P<0.05).In the model group,the expression levels of IL-1β,IL-6 and TNF-α were significantly up-regulated than those in the BC group.关键词
