摘要

Abstract

Objective To study the distribution of D variants among preliminary screening RhD negative blood donors in Liuzhou.Methods 425 blood donors were preliminarily screened by saline method as RhD negative in Liuzhou.These specimens were confirmed as weak D by indirect antiglobulin test.Indirect antiglobulin test negative samples were confnmed as Del and real RhD negative by absorption and elution test.Monoclonal IgM anti-C,c,E,e reagents were used to identify Rh phenotype through saline method.Rh gene frequency,haplotype frequency and Rh phenotype desired value of RhD negative blood donors were calculated by reference found in literature.Hardy-Weinberg goodness of fit was tested.Results Among 425 preliminary screening RhD negative blood donors,99 (23.29%) D variants were identified in all,including 18 (4.24%) weak D and 81(19.05%) Del;326(76.71%) were real RhD negative.The distribution of Rh phenotype was as followed:18 weak D:Ccee 9 (50.00%) >CCee4 (22.22%) >ccEe 3 (16.67%) >CCEe 1 (5.56%) =ccee 1 (5.56%);81 Del:Ccee 47 (58.03%) >CCee 17 (20.99%) >ccee 11 (13.58%) >ccEe 5 (6.17%) >ccEE 1 (1.23%);326 real RhD negative:ccee 199(61.04%) >Ccee 83(25.46%) >CCee 22(6.75%) >ccEe 16(4.91%) >ccEE 3 (0.92%) >CcEe 2(0.61%) >CCEe 1 (0.31%).The observed value and the desired value of Rh phenotype were not statistically significant (P>0.05).The data was analyzed well based on Hardy-Weinberg law.The characteristic of haplotype frequency was cde (0.696 7) > Cde(0.261 6) > cdE(0.041 7) > CdE(0.000 0).The characteristic of gene frequency was c(0.739 5)>C(0.2605),e (0.960 7) >E (0.039 3).Conclusion The detection rate of D variants among preliminary screening RhD negative blood donors in Liuzhou is high,so determining positive weak D and Del blood donors can improve transfusion safety.
