清胰汤联合骨髓间充质干细胞对大鼠急性胰腺炎相关性肺损伤的作用
Effect of Qingyi(清胰) Decoction and Bone Marrow-devived Mesenchymal Stem Cells on Rats with Acute Pancreatitis-associated Lung Injury

Abstract
Objective To observe the therapeutic mechanism of Qingyi decoction and (bone mar-row-devived mesenchymal stem cells, BMSCs) in the rats with acute pancreatitis-associated lung injury (APA-LI). Methods 1.5% sodium deoxycholate was injected retrogradely into the common biliopancreatic duct in rats to make the model of APALI.Fifty rats were randomly divided into sham operated controls (CON) group, acute pancreatitis-associated lung injury (APALI) group,BMSCs (SC) group,Qingyi decoction (QYT) group and Qingyi decoction-BMSCs (QYT-SC) group. After the success of APALI model, mouse model, the tail vein injec-tion of BMSCs suspension was given to the SC group(1 mL/kg),while the Qingyi decoction was given to the QYT group(0.1 g/kg),and the QYT group was also given the Qingyi decoction once after 24 h and 48h. The QYT-SC group was given both BMSCs and Qingyi decoction.All the rats were killed in 7 days after operation and treat-ment.The levels of IL-6,IL-10,MDA and SOD in serum and lung wet/dry (W/D) ratio were measured, compared and analyzed, The distribution of labeled BMSCs in the lung was observed under light microscope. Results 1. The levels of IL-6(141.15±1.35 ng/L),IL-10(114.95±7.24 pg/mL),MDA(54.43±9.77 nmol/mL) in serum and lung wet/dry (W/D) ratio(5.81 ± 0.17) of the APALI group were significantly higher than those of CON group (22.24 ± 1.62 ng/L,35.38 ± 2.81 pg/mL,33.81 ± 4.28 nmol/mL, 4.64 ± 0.27, respectively), and the level of SOD (116.36 ± 8.70 U/mL) was significantly lower than CON group (211.9 ± 8.89 U/mL). The indexes of QYT group, ,SC group and QYT-SC group were better than that of APALI group(P<0.05). The indexes improvement of QYT-SC group was more significant than them in other groups. There were no significant differences in the distribution of labeled BMSCs in different treatment groups. Conclusion Qingyi decoction protects the lungs from injury in many aspects,by inhibiting the production and release of IL-6 and the translocation of bacteria. BMSCs is involved in the pathogenesis of acute pancreatitis, The mobilization of BMSCs can reduce the severity of acute pancreatitis, improve the prognosis. The effect of the combination of the two methods is better in than single treatment.关键词
