Effect of drug-resistant gene mutation sites of Mycobacterium tuberculosis on minimal inhibitory concentration of isoniazid in vitro

Abstract
Objective To investigate the effect of drug-resistant gene mutation sites of Mycobacterium tuberculosis( Mtb) on the minimal inhibitory concentration(MIC) of isoniazid(INH) in vitro.Methods INH-resistant human Mtb strains were screened by Gene chip technology.Then 20 strains with inhA-15(C-T) mutation,and 46 strains with KatG315(G-C) were obtained,and 32 strains without mutation were screened.Bacterial resuscitation was conducted using Lowenstein-Jensen culture.Sensitivity test of INH with gradient concentrations was performed to analyze the MICs of INH for different strains .Results The MIC of INH for the strains with inhA-15 ( C-T) mutation, KatG315(G-C) mutation and non-mutation were(0.375 ±0.079)μg/ml,(5.757 ±4.060)μg/ml,and(0.061 ±0.050)μg/ml,respectively, and there was significant difference among the groups (P<0.05).Low drug-resistance existed in 3 strains without mutation.Of strains with inhA-15(C-T) mutation,drug sensitivity existed only in 1 strain and drug resistance in the others .Drug resistance was found in all of the strains with KatG315(G-C) mutation strains.Conclusion The Mtb strains with inhA-15(C-T) or KatG315(G-C) mutation screened by gene chip are resistant to INH in various degrees .In clinical practice ,the resistance of Mtb to INH can be preliminarily determined according to the drug-resistant result of strains with different mutation sites .关键词
