Effects of PI3K/Akt/FoxO signaling pathway on PRNP expression ofinsulin-like growth factor 1 regulating nerve cells

Abstract
Objective To explore the effects of phosphatidyl inositol 3-kinase (PI3K)/protein kinase B (Akt)/forkhead box O(FoxO) signaling pathway on PRNP expression of insulin-like growth factor 1 (IGF-1) regulating nerve cells PC12.Methods PC12 cells in logarithmic phase were divided into IGF-1 group, experiment group 1, experiment group 2 and the control group.The cells in the IGF-1 group were added with 80 ng/ml IGF-1, cells in the experiment group 1 and experiment group 2 were separately added with 25 and 50 μmol/L LY294002, followed by 80 ng/ml IGF-1 after 30 min 37℃ cultivation.The control group was not given drugs.After 24 h of cultivation, the PRNP mRNA expression of PC12 cells was calculated by real-time fluorescent quantitative PCR in each group.The AKT phosphorylation, AKT phosphorylation (pAKT), FoxO3a and FoxO3a (pFoxO3a) protein expression of PC12 cells was measured by Western blotting in each group.Results The PRNP mRNA and pAKT, pFoxO3a protein expression of the IGF-1 group was higher than that of the control group (all P<0.05).The PRNP mRNA and pAKT, pFoxO3a protein expression of experiment group 1 and experiment group 2 was lower than that of the IGF-1 group (all P<0.05), and the experimental group 2 was lower than the experimental group 1 (P<0.05).Conclusion IGF 1 could regulate PRNP gene expression of PC12 cells through phosphorylation PI3K/Akt/ FoxO signaling pathway proteins Akt and FoxO3a.关键词
