复方苦参注射液对乳腺癌模型大鼠的改善作用
Improvement of Compound Matrine Injection in breast cancer model rat

Abstract
Objective To investigate the effect of Compound Matrine Injection(CMI) on breast cancer model rat.Methods Ninety SPF male SD rats were selected,15 rats were randomly selected for normal group.The rest of the 75 rats were replication breast cancer model by methyl-nitroso-urea intraperitoneal injection.Successful model rats were randomly divided into five groups:model group,control group,large dose experimental group,middle dose experimental group and small dose experimental group,15 rats in each group.The high,medium and low dose experimental groups were given CMI from the second day of the successful model,80,40,20 mL · kg-1 respectively,intraperitoneal injection.The model group was received tamoxifen suspension 40 mL · kg-1 with stomach perfusion.Rats in control group and normal group were given normal saline 4 mL · kg-1 with stomach perfusion,all group were 1 times · d-1.The estradiol (E2) in serum,cell proliferation antigen marker(Ki67),tumor suppressor gene p53 (P53) and programmed cell death 4 (PDCD4) in organization,survivin and PDCD4 in serum were detected by ELISA.Results E2 level in normal group,control group,small dose-,middle dose-,high dose-experimental groups,model group were (36.92 ± 1.28),(70.09 ± 1.91),(64.40 ± 2.77),(40.49 ± 2.17),(40.49 ± 2.39),(49.56 ± 1.27) ng · L-1,respectively;Ki67 level in above groups were(15.72 ±0.76),(70.25 ± 1.59),(65.41 ± 1.33),(21.03 ± 1.50),(21.30 ± 1.10),(40.91 ± 1.85) ng · L-1,respectively;P53 level in above groups were (10.63 ± 0.89),(50.41 ± 1.73),(45.04 ± 2.09),(11.83 ±1.29),(12.18 ± 1.48),(28.37 ± 1.66)ng · L-1,respectively;survivin level in above groups were (5.95 ±0.46),(63.91 ±1.96),(30.85 ± 1.63),(16.96 ±0.53),(16.58 ±0.95),(25.36 ± 1.51)ng · L-1,respectively;PDCD4 level in above groups of organization were (101.06 ±2.49),(31.12 ± 1.37),(35.25 ± 1.13),(79.62 ± 1.73),(79.52 ± 1.45),(56.40 ±2.82)ng · L-1,respectively;PDCD4 level in above groups of serum were (22.00±1.56),(6.93 ±0.62),(7.78 ±0.67),(19.92±1.01),(19.13 ±4.42),(15.34±1.02)ng· L-1,respectively.Compared with control group,serum E2,survivin,organization Ki67,P53 in middle and high dose experimental groups and model group were lower (all P < 0.05).Compared with model group,serum E2,survivin,organization Ki67,P53 in middle and high dose experimental groups were lower (all P < 0.05).Compared with control group,organization and serum PDCD4 in middle and high dose experimental groups and model group were higher (all P < 0.05).Compared with model group,organization and serum PDCD4 in middle and high dose experimental groups were higher (all P < 0.05).Conclusion CMI can improve the levels of PDCD4,P53,survivin and Ki67 in the model rats of breast cancer.关键词
