摘要

Abstract

Objective To detect the expression of PTEN and hypoxia - inducible factor-1α (HIF-1α)in the cancer tissues,para - carcinoma tissuesand and normal tissues of the Mongolian and Han patients with gastric carcinoma,to compare the difference in their expression as well as their interactive effects on the occurrence of gastric carcinoma in the two nationalities. Methods Fifty - seven patients with gastric cancer (including 27 Mongolians and 30 Han people)were selected from the Gastrointestinal Surgery in the Affiliated Hospital of Inner Mongolia Medical University from December 2013 to December 2015. Immunohistochemistry method was used to detect the protein expression of PTEN and HIF-1α in cancer tissues,para -carcinoma tissues and normal tissues of patients with gastric carcinoma,and the mRNA expressions of PTEN and HIF-1α in cancer tissues,para - carcinoma tissues and normal tissues of patients with gastric carcinoma were detected by reverse transcription - polymerase chain reaction (RT - PCR). The expressions of PTEN,HIF-1α protein and mRNA in different tissues of patients with gastric carcinoma were compared and Spearman rank correlation analysis was used to conduct the analysis. Results The positive expression rate of PTEN in the cancer tissues was significantly lower than that in the para -carcinoma tissues and normal tissues of patients with gastric carcinoma (P < 0. 05). There was no significant difference in the positive rate of PTEN in the normal tissues and para - carcinoma tissues between Mongolian and Han patients with gastric carcinoma (P > 0. 05). The positive expression rate of PTEN in the cancer tissues of Mongolian patients with gastric carcinoma was significantly lower than that in the Han patients (P < 0. 05). The relative expression of PTEN mRNA in cancer tissues of Mongolian and Han patients was significantly lower than that in normal tissues and para - carcinoma tissues (P < 0. 05). There was no significant difference in the relative expression of PTEN mRNA in normal tissues and para - carcinoma tissues of Mongolian and Han patients with gastric cancer (P > 0. 05). The relative expression of PTEN mRNA in cancer tissues of Mongolian patients with gastric carcinoma was significantly lower than that of Han patients (P < 0. 05). HIF-1α is only expressed in the cancer tissues of patients with gastric cancer,but not in the normal tissues and para - carcinoma tissues. There was no significant difference in the positive expression rate of HIF-1α in the cancer tissues between Mongolian and Han patients with gastric carcinoma (P > 0. 05). The HIF-1α mRNA electrophoretic bands were only detected in cancer tissues of patients with gastric cancer,but no images were found in the normal tissues and para - carcinoma tissues. There was no significant difference in the relative expression of HIF-1α mRNA in cancer tissues between Mongolian and Han patients with gastric cancer (P > 0. 05). There was a negative correlation between the expression of PTEN and HIF-1α in cancer tissues of Mongolian and Han patients with gastric cancer (r s = - 0. 424, - 0. 408;P < 0. 05). Conclusion A low expression of PTEN is shown in Mongolian and Han patients with gastric cancer and the expression is lower in Mongolian patients with gastric cancer,HIF-1α is only expressed in cancer tissues of Mongolian and Han patients with gastric cancer. The inactivation of PTEN is one of the important causes of gastric cancer,HIF-1α is one of the important factors that promotes tumor growth,and both of them contribute to the occurrence of gastric cancer.
