Effects of adding soybean antigen proteins on serum IgG and IgE levels and expression of intestinal ZO-1 in weaning piglets

Abstract
In order to explore the effect of soybean antigen proteins on allergization and intestinal tight junction Z0-1 protein expression in weaning piglets,seventy 24-days-old weaning piglets were randomly divided into seven groups.The piglets in group A were fed with the basic diet.The piglets in groups B,C and D were fed with basic diets plus 500,2 000 and 5 000 mg/kgβ-conglycinin,respectively.But piglets in groups E,F and G were fed with basic diets plus 500,2 000 and 5 000 mg/kg glycinin,respectively.At age of 24 and 30 days,the serum immunoglobulin levels in each group were measured and determined through ELISA.At age of 30 days,five piglets in each group were slaughtered and the segments of the small intestine were collected for determining the expression of tight junction protein ZO-1 through immunohistochemistry and ZO-1 mRNA relative expression values through qRT-PCR experiment.Statistical analysis showed that adding soybean protein antigens in the piglet diet could increase the serum IgG and IgE levels(P<0.01),but decrease the ZO-1 protein expression and the ZO-1 mRNA expression values in the small intestine.The higher the amount of soybean antigen proteins added,the lower the ZO-1 expression was.The results confirmed that β-conglycinin and glycinin could cause intestinal allergic reactions in piglets,and damage intestinal tight junction.The allergization caused byβ-conglycinin were greater than that by glycinin in piglets.关键词
