Abstract

Objective To separate the total flavonoids and astragaloside from honey-processed Astragalus by a high-efficiency and convenient method and observe its in vitro effect on NO secretion levels.Methods The total flavonoids and astragaloside were separated by Poly-Sery HLB SPE.Lipopolysaccharide was used to induce inflammation model of RAW264.7 cells.The effect of total flavonoids and astragaloside on cell toxicity was detected by MTT.The NO level in culture fluid was detected by Griess method.Results The total flavonoids and astragaloside could be separated very well by Poly-Sery HLB SPE,and the relative contents were 96.59% and 95.06%,respectively.They showed inhibitory acitivity on NO releasing of RAW264.7 induced by lipopolysaccharide,and the effect of astragaloside was stronger than the total flavonoids.The NO secretion level of the model control,total flavonoids and astragaloside were 5.28 μmol/L,2.31 μmol/L and 2.01 μmol/L,respectively.Conclusion Poly-Sery HLB SPE is a highly efficient and convenient method for separating the total flavonoids and astragaloside of honey-processed Astragalus.They have in vitro anti-inflammatory activity.
