摘要

Abstract

Objective To study the antioxidant and anti-aging effect of Dendrobium officinale formula in Caenorhabditis elegans models.Methods The antioxidant activity of D.officinale formula was first determined in vitro using DPPH and hydroxyl free radical scavenging assays.Then food clearance assay was performed to determine the appropriate dosages of D.officinale formula to be used in C.elegans.Survival rate and lifespan of C.elegans were then determined under oxidative stress induced by paraquat and under normal growth conditions,respectively.The antioxidant enzyme activities,reactive oxygen species (ROS) levels and malondialdehyde (MDA) content were measured in C.elegans exposed to paraquat.Results D.officinale formula was capable of scavenging both DPPH and hydroxyl free radicals in vitro.The formula increased the survival rate of C.elegans under oxidative stress induced by 50 mmol/L paraquat and extend the lifespan under normal growth conditions.When C.elegans was exposed to 2 mmol/L paraquat,D.officinale formula was able to reduce ROS level and increase superoxide dismutase (SOD) activity,but the catalase activity and MDA content were not changed.Conclusion D.officinale formula was able to increase the survival rate under oxidative stress and extend the lifespan under normal conditions in C.elegans,possibly through increasing SOD activity.
